Aggressive cancer cells shunt carbon away from aerobic respiration toward anaerobic energy production. With this project we present a strategy to identify genes vital for cancer cells in anaerobic tumors while leaving normal cells on aerobic metabolism unaffected.Jeg har vært i permisjon fra stillingen i perioden 01.09.2014 - 31.08.2015. This project description focuses on glioblastoma (GBM) the most common and most malignant primary brain cancer with extremely poor prognosis. The aggressive nature and the diffuse infiltrative growth pattern of these glial tumors pose a major medical challenge. Current standard of care for newly diagnosed GBM includes surgical resection, followed by combined radio- and chemotherapy (temozolomide), yet the median survival rate is still only 12-15 months. Therefore, this disease urgently calls for new innovative treatment strategies. Since GBMs are highly hypoxic and angiogenic, they have been considered as good candidates for anti-angiogenic therapy either by using VEGF antibodies or tyrosine kinase inhibitors against VEGF receptors. At present, a number of clinical trials have been conducted either as anti-vascular monotherapy or in combination with cytotoxic agents. Unfortunately, in the clinic, anti-angiogenic therapy for brain cancer has not yet met initial expectations despite high radiological response rates after treatment. In a series of preclinical studies using intracranial highly relevant patient-derived xenograft models, we have identified a mechanism that may explain why tumors do not respond to anti-vascular therapy. We have shown that tumor cell resistance to anti-vascular therapy for human GBMs is caused by a metabolic switch towards glycolysis, as indicated by an up-regulation of the transcription factor HIF-1 (hypoxia inducible factor-1) and several metabolites associated with the glycolytic pathway (e.g. lactate; Keunen O, 2011). During this project, using in vitro and in vivo GBM model systems and patient material, we will find potential metabolic targets which may ultimately result in combinatorial treatment principles involving targeting angiogenesis and metabolic pathways in human GBMs.
Bruk av anti-psykotikum som kombinasjonsterapi med Temozolomid i behandling av glioblastoma multiforme.Vi er ute etter å finne en bedring i behandling av den svært aggressive kreftformen glioblastoma multiforme. Pasientene som får den aggressive hjernekreften lever i gjennomsnitt i 15 måneder til tross for å ha fått kreftbehandling.
I dette arbeidet viser vi at en kombinasjon av anti-psykotikum og Temozolomid inhiberer tumor vekst i et in vivo modell system.

Glioblastoma multiforme (GBM) is highly resistant to therapy and carries a dismal prognosis. Standard of care includes surgery followed by radiotherapy (RT) with concurrent and adjuvant chemotherapy with temozolomide (TMZ). The addition of TMZ to RT increases the overall survival by approximately 3 months compared to RT alone, but there are virtually no long-term survivors. We have identified small-molecule sensitizers to TMZ in GBM by integrating a genome-wide lentiviral shRNA library screen with the Connectivity Map database. Individual silencing of 292 genes lead to significantly reduced cell growth in the treated population while silencing of 414 genes increased cellular viability compared to the control group. The Connectivity Map database was then interrogated using these results to identify small-molecule inhibitors that would induce analogous changes in gene expression. This integrated high-throughput approach identified a broad range of compounds with potential cytotoxic activity. Upon validation in low-throughput cytotoxicity assays, we found these compounds effective as single-agent therapies, but also synergistic with concomitant TMZ therapy. Several of these compounds have previously been implicated in anti-cancer therapy, but not as synthetic lethal partners of chemotherapeutic drugs.
We therefore explored the therapeutic potential of these compounds and concomitant TMZ in a clinically relevant intracranial GBM animal model.
We have developed a highly relevant human GBM model which reflects the growth pattern of human tumors. This model has been extensively used in our laboratory and is highly characterized.
Glioblastoma multiforme er en veldig aggressiv form for hjernekreft der pasientene til tross for behandling kun lever i gjennomsnittlig 14 måneder. Vårt mål er å finne komponenter i humane celler som svikter ved utivkling av glioblastoma som kan brukes i terapi mot denne kreftformen.Vi har benyttet oss av lentiviral teknologi til å nedregulere dannelsen av over tusen ulike target i cella som alle er kjent for å være involvert i ulike kreft typer.
Vi har gått fram slik:
Først av alt har vi bestilt et shRNA bibliotek som bærer sekvenser som kan nedregulere over tusen humane gener. Hvert av genene er representert med seks ulike sekvenser. Dette biblioteket har vi laget lentivirale virus av der vårt mål har vært at hver viruspartikkel kun skal inneholde en sekvens.
Etter kontroll at virusproduksjonen har vært en suksess og at våre target celler faktisk lot seg transformere, behandlet vi to ulike glioma cellelinjer in vitro.
Forsøket ble satt opp med ubehandlede celler. Ett sett av celler ble dyrket under hypoksi som er tilstand med 1 % oksygen og skal bedre forestille et tumor miljø. I tillegg inkluderte vi celler behandlet med temozolomid som for tiden brukes i terapi mot glioblastoma.
Cellene ble dyrket og etter en viss tid høstet vi cellene.
Cellepelletene ble sendt til selskapet som shRNA biblioteket kom fra for analyse.
Resultatene har gitt oss mange interessante funn og flere target vi er i gang med å studere nærmere. Nå er målet vårt å velge oss ut noen få aktuelle kandidater som skal jobbes videre med i resten av prosjektet.
Det vi også har startet er å se på noen av disse kandidatene i kombinasjon med temozolomid som er eksisterende terapi mot glioblastoma. Det som er hensikten med dette er at en kombinasjonsterapi vil kunne senke mengde nødvendig medisin for pasienter. Denne vil igjen gi færre bivirkninger.
