During 2014, 5 papers have been published and 1 dissertation defended.In the first 3 papers (Siepel et al., 2014; Pereira et al., 2014, Lebedev et al., 2014a), we investigated neural correlates of cognitive decline in Parkinson's disease (PD) and found that these symptoms are not only present at very early stages of the disease, but also associated with structural and functional changes within the fronto-subcortical and temporo-parietal circuits. In addition, we found evidences suggesting that nigrostriatal dopamine deficiency contributes to PD-related executive impairment. In the fourth paper (Lebedev et al., 2014b), we investigated automated image-based diagnosis of Alzheimer's disease (AD) and found that it is possible to develop tools that would not only demonstrate very high disease detection and prediction accuracy, but also with a good between-cohort robustness, so that a model trained on one large-scale dataset would demonstrate equivalent performance on another sample if the MRI protocols are carefully aligned. Finally, in our fifth paper (Lebedeva et al., 2014) we found that depressive symptoms in AD patients are associated with cortical thinning in temporal and parietal regions and that tau-protein pathology in these areas may contribute to their development in AD. Dissertation “Cognitive impairment in neurodegenerative diseases: insights from computational neuroimaging” was successfully defended on 2014/09/02 (https://bora.uib.no/handle/1956/8473). Future Plans: Establishing Norwegian-Swedish collaboration in the DDI project under the main supervision by Dag Aarsland and Tormod Flatby (Lebedev is currently having a postdoctoral position at Aging Research Center and 20%-position at Centre for Age-Related Medicine, Stavanger University Hospital)

The primary aim of the project is to clarify complex relationships between depression and dementia in the elderly.Summary for 2013: The Helse Vest 2013 funding was extremely helpful for our research project. During this year we managed to establish international collaboration network (Norway, Sweden, UK), prepared 4 papers (2 in 2013, 2 are already published in January 2014), presented our results at the international conferences (EPA-2013, ADPD-2013 + two abstracts are submitted to the MDS-2014 and EPA-2014 congresses), got the access to 3 large-scale databases of Alzheimer's and Parkinson's disease (ADNI, AddNeuroMed, PPMI) and joined the Dementia Disease Initiation (DDI) project. Two other papers are about to be submitted to the journals. Apart from this, the PhD candidate completed his training and results dissemination components, won the short-term fellowship at UCLA, Advanced Neuroimaging Summer Program 2013 (Los Angeles, CA, USA). He is currently working on his thesis (expected submission date: February 2014). Key Findings: - Depression in mild Alzheimer's (AD) and Lewy Body (LBD) dementia is associated with cortical thinning in prefrontal and temporal areas; - Antidepressant use in patients with mild AD and LBD is associated with parahippocampal thinning; - Tau-protein pathology in temporal areas may contribute to the development of depressive symptoms in AD; - Patients with Major Depressive Disorder (MDD) demonstrate abnormally increased anterior hippocampal response to frustration, suggesting that hippocampal-neocortical network impairment may contribute to decreased frustration tolerance associated with MDD; Future Plans: - Joining the PRODE project, as a part of our collaboration with Trondheim and Stockholm (Karolinska Institutet); - Participation in the DDI project (development of image-based biomarkers for early detection and prediction of dementia); - PhD thesis defence.
